There are many nice towns and cities in Czech Republic. The answer to your question depends on your personal interest. In case you just need some ideas - from my point of view of course - here are some hints: Plzen, Ceske Budejovice (both are famous for local brewery); Karlovy Vary (small, but high quality spa resort); Cesky Krumov, (historical sites), Hluboká na Vltavou (its castle is like from fairytales and the nearby zoo at the lake is worth to visit also), Brno (second largest with a nice cathedral and downtown - location for Moto GP). Maybe 2 days in prague, you must go to Cesky Krumlov and Karlovy Vary...
